Biological Models and Hypothesis
A Common Biological Framework?
Research suggests that the thirty-six fast growing diseases and conditions share a common biological profile. The following six biological factors, which are all triggered by the 7th – a single molecule - peroxynitrite – emerge as common bio-markers in these germless diseases. Is peroxynitrite a cause or simply an artifact of chronic disease? Is there a system of biological factors that acts as a framework, which underlies the initiation and perpetuation of chronic disease?
Scientists, Educators and Students
You are invited to collaborate, propose, hypothesize and debate the biological origins and commonality across these fast-growing chronic diseases, which have suddenly experienced exponential growth. The science suggests possible root causes. Or is it simply illusion? The Search for the Cause invites you to share articles that can be read and understood by the general public. We're all in this together. Everyone is touched by this health crisis. Include your links to your research, graphical presentations and support of your research.
This is your platform to communicate with policy makers, funders, clinicians, sufferers of chronic disease and the world.
HINT: Landmark NIH Paper links a single molecule with more than 60 Diseases
In their NIH paper, Pacher et al. build the case for how a complex series of biochemical disruptions, triggered by peroxynitrite (ONOO), sets in motion a lethal combination of oxidative and nitrative stress. Free radicals and nitrative agents like nitrotyrosine disrupt a broad array of biological systems, which, in turn, leads to a self-reinforcing vicious cycle of chronic inflammation, mitochondrial dysfunction and autonomic dysfunction.
The expected outcome predicts disequilibrium, biological chaos and the emergence of disease. Here is a graphic showing the key elements of the chronic disease state, all triggered by peroxynitrite and shared across all diseases tracked in this paper.
- Oxidative Stress (free-radical imbalances)
- Mitochondria Dysfunction (energy center of all cells impacts)
- Chronic inflammation
- Nitrative (NOX) Stress
- Autonomic Dysfunction (AD)
- Epithelial Dysfunction (ED)
- Elevated levels of Peroxynitrite

What is your hypothesis or theory about the root cause(s) of chronic disease? Do biological frameworks predict the onset of chronic disease?
